The 1910 Giro d'Italia was the second edition of the race. It started in Milan on 18 May 1910, and finished in the same city on 6 June, after 10 stages.

Carlo Galetti, classified in second place in the first Giro, won the race with an 18 points margin on Eberardo Pavesi. Previous year's winner Luigi Ganna finished third, 23 points behind Galetti.

One minor change in the race format for this year: points were awarded from 1 point to the winner to 50 points to 50th classified, then all other riders received 51 points.

There were 101 participants, amongst them Jean-Baptiste Dortignacq from France. He was the first non-Italian to win a stage in the Giro, when he and Carlo Galetti formed a breakaway on the second stage to Bologna.

Dortignacq was now considered a danger, and an Italian coalition was formed against him. On stage 3, Galetti, defending champion Luigi Ganna, and Eberardo Pavesi attacked the peloton and the trio rode into Teramo. Galetti won the stage, with Dortignacq finishing 10th. After the 4th stage, Dortignacq got sick, and poisoning was suspected. This resulted in a police investigation, 20 disqualified riders, while Dortignacq had to abandon the race.

The final 6 stages became a battle between Galetti, Ganna and Pavesi. On the final stage to Milan, Galetti, wearing the pink jersey, crashed into a hay wagon mid-way through the race. He managed to get back on his bike and finished 5th in Milan to secure the overall win. Only 20 other riders managed to finished the race.
